EXCLU-SIEVE® Agarose Applications

Agaroses are employed in the preparation of gels used in electrophoresis of smaller  DNA fragments. Major characteristics to be looked for in agarose are strength of the gel, which prevents cracking when handled, and transparency of the gel matrix.

STANDARD AGAROSES 

D-1, in four electroendosmosis measures is our standard multipurpose agarose. It delivers high gel strength thus reducing the risk of cracking when handled as a gel plate. In addition, this product is extremely transparent. Also, because it is made exclusively from seaweed and does not contain any additives, it is almost free of contaminants and inhibitors. A GQT (Genetic Quality Tested) version is available.

HIGH GEL STRENGTH AGAROSE

D-5 (Exclu-Sieve p/n: AHGS) is a linear polymer with high molecular weight and very low sulfate content, producing a gel structure unlike any others. The major characteristics of D-5 are very high gel strength - 3200grs/cm2 at 1,5% - and a higher exclusion limit.

SPECIALIZED AGAROSES

D-2, designed specifically for the production of agarose beads for chromatography and for nucleic acid and protein fraction electrophoresis. Capable of high mechanical resistance and very elastic, D-2 is available in Low Viscosity and Standard Viscosity.
D-3 can give gels with extremely high gel strength (3000 grs/cm2 at 1.5% concentration)  making  it  suitable  for  use  in  the preparation of precast gels.

LOW MELT AGAROSES 

Several varieties are available with melting point temperatures ranging from 65.5ºC to 60ºC at a concentration of 1.5%. Low melt agaroses from The Nest Group offer the highest gel strength in the market: 550 gr/cm2 at 1,5% concentration for LM (Exclu-Sieve p/n: ALM), the most commonly used type, and are very clear upon gelling. Our low melt agaroses have greater sieving properties than standard agaroses. Because of their lower melting temperatures, they are used when recovery without damage is required after electrophoresis. At the same time, lower gelling temperatures (24ºC to 28ºC) permit manipulations such as enzyme digestion, ligation, DNA labelling, etc. while the agarose is in liquid state. SLM and ELM have even lower melting temperatures 25.5ºC and 17.5ºC for specialized applications.
A GQT (Genetic Quality Tested) version of LM agarose is also available.

SMALL SIEVE AGAROSES 

Our MS agaroses are recommended for separating nucleic acids with  less  than  1000  bp  including   PCR  and  DNA  identity (forensic) products. Different products can work efficiently with fragments as small as 9 bp.

According to fragment size, three alternatives are available.

  •  MS-12: 150 bp to 1500 bp, depending on agarose concentration.
  •  MS-8 :  50 bp to 1000 bp, depending on agarose concentration (Exclu-Sieve p/n: AHR)
  •  MS-4:  9 bp to  600 bp, depending on agarose concentration

LM SIEVE is a low melting temperature agarose (65ºC) with very good resolving capacity of DNA fragments smaller then 1000 bp, specially PCR products ranging in size from 200 to 800 basepairs.
